IHC staining with Purified anti-Desmin (clone W23114A) on formalin-fixed paraffin-embedded human heart tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., 10X (Cat. No. 420902). The tissue was incubated without (panel A) and with (panel B) Purified anti-Desmin (clone W23114A), followed by incubation with Alexa Fluor® 647 goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 20X objective. Scale bar: 50 µm -
Cells were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) and permeabilized with 0.5% Triton X-100 for 10 minutes and blocked with 5% FBS for 1 hour at room temperature. HeLa cells (negative control, panel A) and RD cells (positive control, panel B) were then stained with Purified anti-Desmin (clone W23114A), followed by incubation with Alexa Fluor® 647 goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 60X objective. Scale bar: 50 µm -
Whole cell extracts (15 µg total protein) from indicated cell lines and tissue were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-Desmin (clone W23114A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. Desmin is an intermediate filament protein found primarily in muscle cells, where it plays a critical role in maintaining the structural integrity and organization of the cell. Desmin forms a network that connects myofibrils to the sarcolemma and cytoskeleton, providing mechanical support during muscle contraction. This ensures proper alignment and function of the sarcomere, the basic contractile unit of muscle. Desmin also plays a role in muscle mitochondrial dynamics, as loss of Desmin results in defects in mitochondrial morphology, location, and function.

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).
For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with any of the following:- Fixation buffer (Cat. No. 420801) with 0.5 % Triton X-100
- Fixation buffer (Cat. No. 420801) with 100% ice-cold methanol
- 100% ice-cold methanol only
For use in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803), True-Phos™ Perm Buffer (Cat. No. 425401) or True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401). Signal is brightest using True-Nuclear™ Transcription Factor Buffer Set.
