Whole cell lysates (15 µg total protein) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-c-MET (clone A23013D). Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control.     RD cells (negative control, panels A and C) and A-549 cells (positive control, panels B and D) were fixed with Fixation Buffer (Cat. No. 420801), permeabilized with 0.5% Triton X-100, blocked with 5% FBS, and then stained with Purified anti-c-MET (clone A23013D) (panels C and D) or Purified mouse IgG1, κ Isotype Control (Cat. No 400102) (panels A and B), followed by incubation with Alexa Fluor® 647 goat anti-mouse IgG (Cat. No. 405322). Nuclei were counterstained with DAPI (Cat. No. 422801). Description

The c-MET protein, also known as the hepatocyte growth factor receptor (HGFR), is a receptor tyrosine kinase that plays a crucial role in various cellular processes. It is known to be involved in embryogenesis, tissue regeneration, and wound healing. c-MET is activated by binding to its ligand, hepatocyte growth factor (HGF), which triggers a cascade of intracellular signaling pathways such as PI3K/AKT, Ras/MAPK, and Wnt/β-catenin. Regulation of c-MET involves its interaction with other receptor tyrosine kinases like EGFR and RON, as well as its modulation by hypoxia-inducible factors. Aberrant activation of the c-MET/HGF axis, often due to gene mutations, overexpression, or amplification, is associated with the development and progression of various cancers in the liver, lung, and breast. This aberrant activation promotes tumor growth, angiogenesis, and metastasis, making c-MET a significant therapeutic target in oncology.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 2.5 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

For use of this antibody in intracellular flow cytometry (ICFC), we recommend fixation and permeabilization using True-Phos™ Perm Buffer (Cat. No. 425401), True-Nuclear™ Transcription Factor Buffer set (Cat. No. 424401), or Cyto-Fast Fix/Perm Buffer set (Cat. No. 426803).

For use of this antibody in Immunocytochemistry (ICC), we recommend fixation/permeabilization using PFA-Methanol, PFA-Triton X-100, or Methanol only.
