IHC staining with Purified anti-human CD20 (clone W23056F) on formalin-fixed paraffin-embedded human tonsil tissue. Following antigen retrieval using 1X Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704), the tissue was incubated without (panel A) and with (panel B) Purified anti-human CD20 (clone W23056F) followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. The tissue was also co-stained with Purified anti-human/mouse CD3ε (activation epitope) (clone APA1/1) followed by incubation with Alexa Fluor® 594 Goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature (panel C). Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective and merged (panel D). Scale bar: 50 µm -
Daudi cells were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 100% ice-cold methanol for 10 minutes and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained without (panel A) or with Purified anti-human CD20 (clone W23056F) (panel B), followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective and merged. Scale bar: 50 µm -
Human peripheral blood lymphocytes were fixed and permeabilized using True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401) and intracellularly stained with PE anti-human CD19 (clone HIB19) (Cat. No. 302208) and Purified anti-human CD20 (clone W23056F) (left), orPurified anti-rat IgG2a, κ Isotype Control (right) (Cat. No. 400501) followed by Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416).

CD20 is a cell surface protein primarily expressed on B lymphocytes which plays a key role in the regulation of B cell activation, proliferation, and differentiation. Encoded by the gene MS4A1, homo-oligomeric complexes of CD20 form calcium conductive ion channels that modulate signaling pathways necessary for B cell survival and immune responses. CD20 plays a role in B cell activation of downstream Src family kinases such as Lyn, Lck, and Fyn. Because of its selective expression on B cells CD20 has become a target for therapeutic monoclonal antibodies to treat conditions like B cell lymphomas and autoimmune diseases.

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).
For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with either of the following:
-Fixation buffer (Cat. No. 420801) with 0.5 % Triton X-100
-Fixation buffer (Cat. No. 420801) with 100% ice-cold methanol
-100% ice-cold methanol only
For use in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803) or True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401).
This product is not recommended for use in Western Blot.
