    Stability Testing for Recombinant Human B3GAT1. Recombinant human hB3GAT1 was aliquoted in 20mM Tris, 150mM NaCl, pH 7.0 at 0.2 mg/mL and one aliquot was kept at 4°C (Control), and another was frozen and thawed four times (4x Freeze/Thaw). After this procedure, the samples were tested for their ability to inhibit the proliferation of U-118 MG human glioblastoma cells. The ED50 for this effect is 0.8 – 4.8 µg/mL.

Description

The B3GAT1 gene encodes the glucuronosyltransferase P (GlcAT-P) enzyme, a member of the glucuronosyltransferase family, which plays a role in glycosaminoglycan (GAG) biosynthesis. This enzyme facilitates the formation of the GlcAβ1,3-Gal linkage on glycans. Substitution of Phe245 in GlcAT-P with alanine reduces the enzyme's binding affinity to its ligand. B3GAT1 is also involved in the production of the cell surface carbohydrate epitope HNK-1/CD57. The HNK-1 epitope is found as a residue on several glycoproteins and glycolipids. HNK-1-positive memory T cells exhibit impaired proliferative capacity, suggesting that this epitope may serve as a marker of immune cell senescence. The HNK-1 epitope is also detected in various cell adhesion molecules (e.g., NCAM, L1) and extracellular proteins (e.g., tenascin-R, phosphacan). B3GAT1-induced HNK-1/CD57 expression acts as a biomarker for neurological conditions such as Alzheimer's disease and psychosis. Additionally, elevated levels of B3GAT1 have been observed in human brain tissue. When combined with existing markers, HNK-1 provides a more robust diagnostic tool for neuroendocrine neoplasms (NEN). In an astrocytic tumor cell model, overexpression of B3GAT1 leads to HNK-1-positive cells. These cells exhibit ERK 1/2 inactivation, resulting in reduced proliferation in glioma cell models and smaller tumor growth in animal models compared to control groups.

Molecular Mass
The 316 amino acid recombinant protein has a predicted molecular mass of approximately 36.26 kD. The DTT-reduced and non-reduced proteins migrate at approximately 35 -62 kD and 80 – 115 kD by SDS-PAGE, respectively. The predicted N-terminal amino acid is His.

Concentration
25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin the vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 μg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.

Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.

Does specific activity of a recombinant protein vary between lots?

Specific activity will vary for each lot and for the type of experiment that is done to validate it, but all passed lots will have activity within the established ED50 range for the product and we guarantee that our products will have lot-to-lot consistency.
