Description

Nectin-1, also known as poliovirus receptor related 1 protein (PRR1) and CD111, is a type I transmembrane glycoprotein with three extracellular immunoglobulin domains, a single transmembrane helix, and a cytoplasmic tail. Nectin-1 is part of the immunoglobulin (Ig) gene superfamily and a member of the Nectin family: Nectin-1, Nectin-2, Nectin-3, and Nectin-4. Nectin-1 plays a key role in arbitrating cellular entry of the herpes simplex virus by interacting with the viral glycoprotein D (gD). Nectin-1 is broadly expressed in endothelial cells, epithelial cells, neuronal cells, megakaryocytes, and CD34-positive stem cells. Nectin-1 localizes in various junctions such as the chemical synapse of neurons and the Adherens Junctions (AJ) of epithelial tissue. Nectin-1 promotes cell-cell contacts by forming homophilic or heterophilic trans-dimers together with Nectin-3 or Nectin-4; Nectin-1 also binds to afadin, a multi-domain scaffold protein.

Biotinylated recombinant human Nectin-1, amino acid (Gln31-Gly346) (Accession: #Q15223.3), with a linker, C-terminal human IgG1 (Pro100-Lys300) and an -Avi tag was site-specifically biotinylated by enzyme BirA and was expressed in 293E cells.
Molecular Mass
The 568 amino acid recombinant protein has a predicted molecular mass of approximately 63.75 kD. The DTT-reduced and non-reduced proteins migrate at approximately 70 -115 kD and 140 – 240 kD by SDS-PAGE, respectively. The predicted N-terminal amino acid is Gln.
Purity
> 95%, as determined by Coomassie-stained SDS-PAGE.

0.22 µm filtered protein solution is in PBS, pH 7.2
Endotoxin Level
Less than 0.1 EU per µg of cytokine as determined by the LAL method
Concentration
25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin the vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 μg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
Activity
When recombinant human Nectin-3 (Cat. No. 566604) is immobilized at 1 µg/mL, biotinylated recombinant human Nectin-1 binds in a dose-dependent manner with ED50 range of 3.6 – 36 ng/mL. HRP-Avidin (Cat. No. 405103) was used to detect the binding.
