An Expanding Portfolio of Research Tools for Interleukins and their Receptors

Upon recognizing their ligands, interleukin receptors initiate a cascade of signals and a variety of immunological responses. To aid researchers, BioLegend continuously improves its catalog of interleukin receptor antibodies, offering a large selection of specificities in a wide array of fluorophore conjugates. The common gamma chain (IL-2Rγ) can pair with IL-21R to mediate Th17-related inflammation or with the IL-4R to promote Th2 function and IgE production. IL-13 utilizes IL-4R and IL-13Rα1 as its receptor. IL-13Rα2 can compete for IL-13 and limit its positive influence on allergy and Th2 cells. IL-17RB and IL-17RA bind to IL-25 (IL-17E) and also mediate allergic responses while expanding eosinophils. Petersen et al. investigated the importance of IL-17RB in generating TM2 cells and allergic responses.

IL-17RB deficiency can reportedly reduce asthma pathology induced by allergens, while polymorphisms in IL-17RB can increase risk for asthma. Petersen et al. studied IL-25's potent Type 2 proinflammatory functions by instilling it in the airway of mice. IL-4 and IL-13 production was upregulated in a novel population, the Type 2 Myeloid (T2M) cells. T2M cells were characterized as granulocytic and most closely related to eosinophils, with the cells expressing a phenotype of IL-17RB+ Gr1mid CD11b+. T2M cells could even introduce IL-25-mediated lung pathology when adoptively transferred into IL-17rb-/- mice. This work sheds light on the role of IL-25/IL-17RB in chronic allergic airway diseases.
