Leukemia Inhibitory Factor and New Recombinant Proteins

Of our new recombinants, we're featuring LIF (Leukemia Inhibitory Factor), which is a pleiotropic cytokine of the IL-6 family. LIF is thought to be essential for the completion of human pregnancy. It was originally discovered in mice, where it limited proliferation of myeloid leukemic cells and instead, caused macrophage differentiation. LIF is also known to control uterine receptivity for blastocyst implantation and trophoblast development. LIF knockout mice are incapable of properly implanting a blastocyst on the uterine wall. In addition, LIF gene mutations have been associated with infertile women. Suman et al. reviewed the link between LIF, STAT3, and embryo development.

LIF is expressed by endometrial glands and epithelium. Both the blastocyst and endometrial epithelium can express LIFR (LIF Receptor). At this time, the endometrium will form pinopodes (ectoplasmic protrusions; not shown in figure) to latch onto the blastocyst. Pinopodes increase expression of LIFR and integrins to bind to the blastocyst. Other factors on the endometrium like HB-EGF (heparin-binding EGF-like growth factor) upregulate expression of integrin ?5?3 on trophoblast cells, which form the outer layer of the blastocyst. A combination of these factors allows the blastocyst to be properly implanted on the endometrium. LIF signaling on blastocysts activates and phosphorylates STAT3, which leads to a variety of functions, including cytokine secretion, differentiation, proliferation, and the invasion (or successful implantation) of the blastocyst into the endometrium.
